چکیده

From replacement studies (1) and inhibition analysis (2) with microorganisms requiring folic acid, it has been inferred that this vitamin is concerned in the biosynthesis of purines and of thymine. More directly, folic acid has been shown to be involved in nucleic acid synthesis in Lactobacillus casei (3,4). According to Buchanan and Schulman (5), the formation of purine derivatives would seem to require a folic acid-activated enzyme. The observed effectiveness of uracil in large doses in pernicious anemia in relapse has been interpreted to mean that it may be a precursor of thymine (6). Interchangeability of certain pyrimidines in the nutrition of microorganisms is known (7-9). The present study relates to the ability of several strains of Bacillus subtilis to convert uracil to thymine and to the probable role of folic acid and vitamin Blz in this transformation.

On the mechanism of thymineless death in Bacillus subtilis.

Thymineless death (TD), the loss by a thymine-requiring auxotroph of the ability to multiply after a period of thymine starvation, was first described1 in Escherichia coli 15T-, and was attributed to "unbalanced growth," the synthesis of RNA and protein in the absence of DNA synthesis. This phenomenon has been investigated in thymineless auxotrophs of a number of bacterial species: E. coli 15Ta...

BaciZZus subtilis Bacteriophage PBS2-induced DNA Polymerase

The DNA polymerase induced by Bacillus subtilis bacteriophage PBS2 (whose DNA contains uracil instead of thymine) has been purified and characterized for its specificity. The enzyme requires a high ionic strength for optimal stability and activity and is sensitive to various anions and to sulfhydryl reagents. Both dUTP and dlTP are incorporated efficiently as substrates and are competitive inhi...

Uracil incorporation into nascent DNA of thymine-requiring mutant of Bacillus subtilis 168.

A thymine-requiring mutant of Bacillus subtilis strain 168 accumulates short DNA chains after brief pulses with [(3)H]thymidine. Reversion of the thy mutation to thy(+) abolishes the accumulation of short DNA chains, suggesting that the accumulation is related to the thy mutation. The reason for this accumulation has been further investigated by analysis of a mutant with a defective uracil-DNA ...
